Etude De L’effet Hépato-préventif De L’extrait Aqueux D’astragalus Gombiformis Pomel. Contre La Toxicité Induite Par La Doxorubicine

Résumé: L’objectif principal de cette étude est l’évaluation de l’effet préventif de la plante médicinale Astragalus gombiformis Pomel.sur la toxicité hépatique induite par la doxorubicine, après avoir confirmé qu’il est sans effet toxique (DL50 supérieure 5g/Kg de poids corporel). Les souris de souche Swiss albinos ont été soumises à un traitement oral de l'extrait aqueux (2g/kg Pc) contre l'hépatotoxicité induite par une injection intrapéritonéale unique de doxorubicine (15 mg/kg). L'hépatotoxicité a été évaluée en mesurant le taux sérique (AST et ALT), les activités des enzymes antioxydants (GPx et MDA) et des études histologiques. D’après les résultats, on constate que EAAg (200mg/kg Pc) peut protéger le foie contre la toxicité provoqué par la doxorubicine en augmentant le niveau des enzymes antioxydants GPx, et l’inhibition de la peroxydation lipidique ce qui empêche la libération des transaminases dans le sang. Ces résultats ont été confirmés par les études histologiques. Mots clés : Astragalus gombiformis Pomel. , doxorubicine, hépatotoxicité, stress oxydant Abstract The main objective of this study was evaluating in vivo the preventive effect of the aqueous extract of the plant Astragalus gombiformis Pomel. against the hepatotoxicity induced by doxorubicin ,after confirming that it has not toxic effect (LD50 greater than 5g /Kg of body weight ). Swiss albinos mouses were subjected to oral treatment by gavages with the aqueous extract (2g / kg Pc) against hepatotoxicity induced by single intra-peritoneal injection of doxorubicine (15 mg/kg).Hepatotoxicity was assessed by measuring the level of serum (AST, ALT), the activities of antioxidants enzymes (GPx and the level of MDA) and histological study. According to the results, it appears that the aqueous extract of Astragalus gombiformis Pomel. (200mg/kg Pc) can protect the liver against the acute toxicity of doxorubicin by increasing the level of antioxidant enzymes GPx and the inhibition of lipid peroxidation which prevents the release of transaminases in the blood.
